My understanding is that as long as the Pi is connected to the supply, the 3.3v is always on, even in standby.
5v is always on. 3.3v comes from the pmic. If "all outputs are switched off" there cannot be 3.3v.
Why not just try it and see? It's not like its going to cause any damage.
